Abstract: In this experiment we are trying to figure out if different PH solution affects the activity of an enzyme, does it affect the enzyme? Or does it not affect the enzyme at all? We see that the PH 7 solution has the highest absorbing value, showing use the optimum PH while the other PH solutions are less. Showing us that PH does have an effect on enzymes but, also showing that PH 7 is the optimum PH, meaning that the reaction at the fastest rate than at any other pH level. Introduction: Does PH affect an enzyme’s activity? There are many things that that can affect an enzyme in different ways. Such as temperature affecting the reaction rate of an enzyme. We then added 1 ml of enzyme and 5 ml of distilled water. We need to make sure to cover the test tube with parafilm and made sure to mix the contents well. The spec 20 wavelengths had to be set to 540nm, we had to adjust it. We then used the spec 20 to set the absorbance read to infinity with the left knob and made sure to wipe the test tubes, any prints/oils could affect the experiment. Now using the right knob of the spec 20 we set the absorbance reading to 0, then removed the blank from the spec 20 and save it for later on in the experiment. We then obtained 4 test tubes labing them 1-4. Add 3 ml of distilled water in all 4 test tubes and add 3 ml of different PH solutions (5,7,9) in each test tube except for test tube numbered 4 and mix contents well. Then we added 1 ml of Catecholase in the first 3 test tubes contain PH and add 1 ml of Catechol in all test tubes, and be sure to put a parafilm on all test tubes.
