BB1802 1618883 BB1802 Microscopy coursework- Introduction- Figure 1: shows the different parts of a light microscope. Light microscopes are microscopes used to examine objects using light. Light microscopy is one of the least invasive techniques used to access information from various biological scales in living cells. The combination of molecular biology and imaging provides a bottom-up tool for direct insight into how molecular processes work on a cellular scale (Antony et al.,2013). Gram-negative bacteria are surrounded by a thin peptidoglycan cell wall, which itself is surrounded by an outer membrane containing lipopolysaccharide. Gram-positive bacteria lack an outer membrane but are surrounded by layers of peptidoglycan many times thicker than is found in the Gram-negatives (J.Silhavy,Kahne and Walker,2010). Which is why after treatment with a decolorizer the lipid layer from the Gram-negative cells is dissolved whereas, in contrast, the solvent dehydrates the thicker Gram-positive cell walls closing the pores and as a result diffusion cannot happen so the bacteria remain stained (Personal.psu.edu,2017) as seen in image 4 the rod-shaped bacteria have a purple-violet …show more content…
