Abstract
Combination therapy can be one of the vital ways of treatment for cancer. However, its current application with some chemodrugs has have certain limitations per se, due their short half-lives and low tumor application. Nano based drugs in this case show a great assurance with providing the superior treatment benefits. Following essay shows the general idea of Nanoparticles based drug delivery in combination, for therapeutic application rather than codelivery of single Nano drug carriers. Also, various strategies like, targeting and tumor therapy with dual nanoparticles in combination. Finally, recent advances; limitations and future selection of therapeutic agents; targeting efficiency and the gap between the preclinical and clinical outcome is discussed.

Even, by the treatment with cytotoxic drugs or molecular targeted treatment incurs huge challenges like drug resistance, insufficient curative effects and tumor relapse and so forth. However, combination therapy with multiple therapeutic agent act on multiple sites, or increase the tumor sensitivity to multiple one another creates enormous potential for cancer treatment[1]. Despite many possible advantages, the current clinal combination therapeutics have many disadvantages. The possible cumulative toxicity may lead to intolerance in patients[2]. Further, the poor water solubility of active pharmaceutical ingredients(API), short elimination half-life of API reduces duration of efficient treatment. Also, variation in pharmacokinetics(PK) and bio-distribution of anti-cancer agents makes it unable to attain required stoichiometric ratio at tumor site or exert synergistic tumor inhibition.
