1 1STB1013 Cell Biology Laboratory Manual: Scanning Electron Microscopy (SEM) Introduction Welcome to the Cell Biology laboratory demonstration on Scanning Electron Microscopy (SEM). In this session, you will have the opportunity to explore the principles, techniques, and applications of SEM in cell biology research. This manual will guide you through the process of the laboratory demonstration and help you prepare a comprehensive lab report that showcases your understanding of the topic and your abilities in leadership, autonomy, and responsibility. Objectives 1.To understand the principles and working mechanisms of SEM. 2.To observe and analyze biological specimens using SEM. 3.To learn the importance of leadership, autonomy, and responsibility in a laboratory setting. Equipment and Materials •Scanning Electron Microscope (SEM) •Biological specimens (provided by the instructor) •Safety goggles •Lab coats •Gloves •Notebooks and pens Safety Precautions 1.Always wear safety goggles, lab coats, and gloves when working in the laboratory. 2.Follow all safety instructions provided by the instructor. 3.Avoid touching your face or eyes while working with specimens. 4.Notify the instructor immediately in case of any accidents or spills. Laboratory Demonstration: Scanning Electron Microscopy (SEM) 1.SEM Principle and Operation:•The demonstrator will provide a brief overview of SEM principles and how the microscope operates. •Pay close attention to the instructor's explanations and ask questions if needed. 2.Sample Preparation:•Observe the demonstrator's demonstration of specimen preparation for SEM. •Note the steps involved in fixing, dehydrating, and coating the sample. 3.SEM Imaging:•Work in groups of 5 students. •Each group will have the opportunity to load a prepared specimen into the SEM.
2 2•Observe the SEM images of the specimen and discuss your observations with your group. 4.Data Collection:•Take notes on the SEM images, magnification settings, and any interesting features you observe. •Discuss with your group members and gather data for your lab report.
3 3Lab Report Instructions PART 1: WRITTEN REPORT 1.Your lab report should be typed in Font 12 and organized neatly. 2.Include a cover page with the following information: •Title: "SEM Laboratory Report" •Your name and the names of your group members. •Date of the laboratory demonstration. 3.The report should be structured as follows: •Briefly introduce the purpose and objectives of the laboratory demonstration. 4.SEM Section •Describe the principles of SEM. •Discuss the sample preparation process. •Present SEM images and analyze the observed features. 5.Discussion •Comparing SEM with Other Microscopy Techniques (10%) •Discuss the significance of the observed features in the context of cell biology. 6.Conclusion •Summarize the key findings and their relevance. 7.References •Cite any sources you used to prepare your report.
