Analysis of Unknown Samples Through Staining and Chemical Testing (Unknown 8) Abstract: Identifying bacteria by culture is possible due to various tests ranging from staining techniques to biochemical testing. Bacteria have a wide range of variances. To help identify unknown bacteria, gram staining, selective and differential media, analytical profile index (API), and BLAST tests were performed. It resulted in gram negative, rod shaped, lactose fermenting bacteria. The most likely bacterium it could be is Klebsiella pneumoniae. The morphology of the bacteria is a rod shape. Through this experiment, it can be concluded that the bacteria is a Gram negative species. The selective and differential plates were left to incubate and after viewing the results, there was no growth on either of the plates. On the MAC plate, there is a bit of discoloration of the phenol red pH indicator. Though there was a bit of discoloration, it is not a strong enough indication that the bacteria reacted with the agar because there was no growth. The MSA plate also showed discoloration throughout the plate and shows a stronger indication in comparison to the MAC plate shown in Figure 2. Though there was no growth on the plate, the strong discoloration indicates that the bacteria reacted with the sodium chloride in the agar. The API test resulted in mostly negative results in amino acid catabolism and mostly positive in fermentation.
