Aquatic Microbiology Lab Report
Metagenomics is the study of genetic material recovered directly from environmental samples. It's shotgun sequencing of microbial community DNA provides an in depth look at genotypic richness within populations. This process gives microbial biologists great insight into the diversity that microbes posse and can provide us with many novel genes that have never been seen before. This allows us to look into the many different types of metabolisms that can occur in life. Big limitation is we don't know if they are actually doing something in the environment. Gives nice big picture but not specifics.
On February 10th, we extracted DNA collected from multiple locations across the island. This included paradise falls, oyster water, beach sand, and wastewater. In order to do extract the DNA from the four locations, we followed a procedure that included

We next added Solution C1 aided in the cell lysis that was started in the previous step, followed by a 10 minute centrifuge. We then added solution C2 to our solutions which precipitated non-DNA material, increasing the purity of the DNA we planned to extract. We then transferred the supernatant (while avoiding the pellet) to a collection tube to remove the precipitated pellet that contained non-DNA materials. This was followed by an the addition of C3, which removed additional non-DNA material, to the supernatant, some vortexing, and then incubation. We again transferred the resulting supernatants to another clean tube to
