SUMMARY The purpose of this experiment is to be able to identify Bacillus megaterium using 16S PCR & Sequencing and also determine whether bacillus megaterium is susceptible or receptive to antibiotics using gram stain. Being able to determine whether bacillus megaterium is susceptible or receptive to antibiotics is critical to medical advancement and biotechnological enhancements. INTRODUCTION AND BACKGROUND Bacillus megaterium is a gram positive, endospore forming, rod shaped bacteria, motile, and it is not capable of butanediol fermentation. It is considered aerobic and one of the first bacteria's genome that has been fully coded. It is found in soil and considered a saprophyte. Bacillus megaterium is Latin for the big beast because it is an enormously large bacteria. Gram positive organisms are susceptible to antibiotics such as penicillin (Introduction to Penicillins, 2012). This is because, gram positive bacteria have a thick layer of peptidoglycan, a sugar and peptide coating that gives a cell its shape and helps it stay intact (Wiley, 2004). The original antibiotic, penicillin, and the likes are used to treat infections caused by bacteria that are gram positive. Based on the scope of the experiment and the materials needed, a budget estimate of $1,000.00 would be needed to buy reagents and materials for this experiment. If this experiment is funded and performed, its benefits will be enormous.
