The purpose of this experiment was to insert the plasmid glow green into the bacteria with a gene of interest to produce the protein that make the bacteria glow green along with the presence of arabinose and the presence of ampicillin. Many scientists are experimenting different kind of genes that can inserted into the organism for survival. The technique of transformation was used in this experiment to give the organism a new trait that they did not possess in their life. In this experiment, the bacteria were added to four plates with certain conditions such as the existence of plasmid, ampicillin, and arabinose to see whether the bacteria grow and glow green. With a new gene expresses, it can lead to genetic diversity. In daily basis experiments, scientists are introducing a new gene that can amplify the plasmid and produce large quantities of a new characteristic. This experiment builds a lot of beneficial for medicine and biotechnology. This technique called Transformation was introduced when a gene from the jellyfish was extracted and placed into the plasmid that encodes for Green Fluorescent Protein to produce the gene that give the characteristic of glowing green. This process involves many factors that will be tested to see whether the bacteria has the plasmid and how the bacteria grow and survives with a new trait. The principle of Transformation on E.Coli is that most bacteria will survive with transformed cell containing the plasmid and antibiotic resistance. Survival bacterial with the plasmid will then glow green with three different genes. Bacteria or E.coli played a significant role in this experiment because it contains the DNA that will inherit new genetic material from the plasmid. Plasmid glow green or pGLO also played a significant role because it was inserted in the bacteria to transfer the gene into the bacterial DNA. This experiment was tested on two conditions: +pGLO and -pGLO. +pGLO has the plasmid and -pGLO does not have the plasmid. Along with those conditions, ampicillin is added in the agar plate to destroy the non-transformed cells because scientist want to select only the transformed bacterial cells and the experiment is efficient at ten percent. To resist the antibiotic, the bacteria needs Beta lactamase that provide the enzyme. This allows the bacterial to break down the antibiotic and grow and with the plasmid. In addition, the bacteria need arabinose that help activate the protein that allows to glow green in the transformed bacteria cells. To let
