In the stickleback fish found in Big Lake (seen in slot #4 and #5) are about 1400 base pairs long. By using PCR, we were successfully able to isolate the Pitx1 enhancer region in the marine control, and both stickleback fish in Big Lake. The sterile water used in the experiment was not contaminated, as seen in slot #6 of the gel electrophoresis, which means the results of the experiment should be accurate and without any contamination. There is a deletion in the Pitx1 enhancer region of the stickleback fish found in Big Lake, since the Pitx1 enhancer region is approximately 117 base pairs shorter than the marine
The results of the gel electrophoresis are summarized in Figure 2. On the gel, both populations of fish were run and analyzed for their heterozygosity. Following the conclusion of the electrophoresis, the gel was analyzed to determine how many different alleles were present at the SFMSTR5 loci. The results of the analysis are shown in Figure 2. The gel showed that in population 1, there are three different alleles at the SFMSTR5 loci and that a majority of the fish in this population are heterozygous at this locus.
The experiment consisted of two sub-experiments that when synthesized together, successfully answered our main question. During the summer and winter months, it’s considered as the harvest season as various salmon species, especially Sockeye and Chinook, would swim return to the tahltan territory to spawn. The Tahltan would set up along “the mid-Stikine, upper-Nass and upper Skeena basins with gill-nets ready for the harvest.” (Rescan. 2012) The fish caught during this season are smoke-dried and are used to feed their families throughout the season.
The increasing temperature in the Columbia and Snake River Basin will provide less of a habitat range for the cold water species, sockeye salmon. The reason being that if the salmon were to attempt to inhabit these warmer waters the individuals that are unable to physiologically acclimate to these new temperature conditions will die; thus decreasing the relative abundance of the sockeye salmon’s population in these areas.
