Omalizumab
Omalizumab is a humanized monoclonal antibody used for the treatment of persistent allergic asthma. In general, allergic reactions are triggered by IgE present in human immune system. Allergies predominately occur on skin surface, in gut, in respiratory tract, or any other sites of parasitic invasion. The persistent allergic asthma is the allergic reaction at the respiratory tract that initiates inflammation, bronchoconstriction and mucus secretion. Omalizumab binds to the Cε3 domain of IgE and forms hexamer complex that inhibits the immune response by preventing IgE from attaching to mast cells and other effector cells, thus down-regulating the FcεRI receptors (see Appendix for more information).

As a result, FDA has added information about these potential risks to the drug label. The review also found that there is no difference in the rates of cancer between those patients being treated with Xolair and those who were not being treated with Xolair. However, due to limitations of 5-year study in analyzing the cancer rates associated with Xolair, FDA is currently not in the state of ruling out the potential risk of cancer with this drug.
